Working on a quote for a routed sign. customer wants 5'x10' hdu... What thickness should be used on something this large? is 1.5" thick enough... gonna be 1 sided and I am plan to back with aluminum for strength.

Depends on how deep you're going to be routing, but in my opinion, that's fine. What will it be attached to? It will want to bend a bit with daytime temperature changes, and can you get 10 x 5 ft HDU in one piece where you are?
Here's one we did last week - 1/8" alupanel backing 3/8" narrower all round, 1 1/4" HDU for the whole shape, and 1" HDU glued on the ovals and 'hand' panels, to give extra depth to the body. There's a steel frame behind that, too.

I was gonna use like 1/8" Polymetal or something and 1.5" thick all one piece and only route 1/4" depth.. Gonna be mounted between 2 stone columns

Polymetal has practically no strength at all. It's fine for small signs. The aluminum skin is very thin and the core is just plastic. There have been several discussions on this topic on this board. You could do a search for HDU signs and see good examples of mounting HDU between two posts.
